چکیده

In this short account, we have focused on the methodology for the single-molecule detection of reactive oxygen species (ROS), such as hydroxyl radical (HO) and singlet oxygen (O2), using a total internal reflection fluorescence microscopy (TIRFM). A successful application using the single-molecule fluorescence techniques for the investigation of the TiO2 photocatalytic oxidation reactions is briefly demonstrated.

Use of tapered amplifier diode laser for biological-friendly high-resolution optical trapping.

A 1064 nm laser is commonly used for biological optical trapping. However, it has the problem of generating reactive oxygen species in the presence of a sensitizer, which leads to photo damage in biological samples. Here we constructed optical tweezers using a tapered amplifier diode laser that operates at 830 nm. Compared to a 1064 nm laser, this laser is friendly to live cells, eliminates pho...

Fluorescence probes used for detection of reactive oxygen species.

Endogenously produced pro-oxidant reactive species are essential to life, being involved in several biological functions. However, when overproduced (e.g. due to exogenous stimulation), or when the levels of antioxidants become severely depleted, these reactive species become highly harmful, causing oxidative stress through the oxidation of biomolecules, leading to cellular damage that may beco...

Single-cell analysis of dihydroartemisinin-induced apoptosis through reactive oxygen species-mediated caspase-8 activation and mitochondrial pathway in ASTC-a-1 cells using fluorescence imaging techniques.

Dihydroartemisinin (DHA), a front-line antimalarial herbal compound, has been shown to possess promising anticancer activity with low toxicity. We have previously reported that DHA induced caspase-3-dependent apoptosis in human lung adenocarcinoma cells. However, the cellular target and molecular mechanism of DHA-induced apoptosis is still poorly defined. We use confocal fluorescence microscopy...

Two-photon fluorescence imaging of intracellular hydrogen peroxide with chemoselective fluorescent probes.

We present the application of two-photon fluorescence (TPF) imaging to monitor intracellular hydrogen peroxide (H₂O₂) production in brain cells. For selective imaging of H₂O₂ over other reactive oxygen species, we employed small-molecule fluorescent probes that utilize a chemoselective boronate deprotection mechanism. Peroxyfluor-6 acetoxymethyl ester detects global cellular H₂O₂ and mitochondr...
